You just need to get the use of those locking tuners sorted out. To get the best from them, you shouldn't have more than one complete turn of string around them. Line the tuner hole up with the neck so a new string can run straight into it. Pull it hand tight, then lock it. You should normally then be able to tune up with 1/2 to 3/4 of a turn. The whole idea of locking tuners is that you don't have 3 or 4 turns of string around the post, as it's these turns that move position when you use the trem and slacken off the string tension, and so the tuning can suffer.
