1+ on having the high E saddle 3/4 toward the nut...or even a touch more.

I'd put it "a touch more" if you are doing string through. Not so much because of intonation issues, but to give you a bit more break angle off the back of the saddle on the low E. 3/4 is also good if you are using a three saddle bridge since the position for the high E and the B strings will be in the same place.

The beauty of the Tele bridge is that the saddles have a huge range of travel, so you don't have to worry about that very much. It's more of an issue on the Tune-o-matic bridges as in Drashkum's build because they have a relatively small range of travel.

The rule of thumb is that no string will need less than the exact scale length, but lower strings may need a bit more.

BTW you don't need to do the measurement from the 12th fret. I think most people measure from the nut to the saddle directly. I have mostly see advice here about measuring from the nut to the saddle...but both approaches work in my experience. The advantage to measuring from the nut is that, since you have the straight edge running the full length it helps in getting the strings aligned over the fretboard and pole pieces.
